D-dimer is a final product of fibrin degradation. Elevated D-dimer levels in blood are considered a reliable marker of pathological coagulation, which underlies the pathogenesis of many cardiovascular diseases. D-dimer testing is widely used to help exclude the diagnosis of deep vein thrombosis (DVT).

Hytest has long offered flagship products for the development of D-dimer assays. We provide different antibody recommendations depending on the assay goal and application platform. All recommended immunoassays are intended for the quantitative determination of D-dimer and high molecular weight fibrin degradation products. All monoclonal antibodies (MAbs) recognize D-dimer in ELISA.

Building on DD255cc, we have recently launched the recombinant version RD15. RD15 is produced recombinantly to secure long-term supply and lot-to-lot consistency, while remaining fully murine in structure. As a result, assay performance and interference characteristics remain comparable to traditional murine clones, simplifying transition and re-validation for IVD manufacturers already using DD255cc, as well as onboarding for new customers. Moreover, we offer RD15 in bulk with shorter lead times and competitive pricing. The recommended antibody pairs for RD15 correspond to those for DD255cc.

*Due to the cross-reactivity of DD4 with fibrinogen, we strongly recommend using it as the detection antibody. In a sandwich immunoassay, plasma must be diluted at least two-fold with 10 mM Tris-HCl, pH 7.5, 1 M NaCl, 0.1 % Tween 20 to avoid nonspecific binding. Each step in the assay should be followed by an incubation and wash: coating with the capture MAb, addition of the sample and addition of the (conjugated) detection MAb.
